<p>Scroll through any longevity podcast and you will hear the same four names: NMN, NR, resveratrol, and metformin. They get described as the molecules that flip your cells back to a younger state, and the bottles can run well over a hundred dollars a month. The promise is enormous, and the marketing is louder than the evidence.</p>
<p>Here is the honest version. Each of these compounds has real science behind it, and most of that science is in mice, not people. Knowing what the research actually shows, and where it stops, is the difference between a smart experiment and an expensive guess.</p>
<h3>What These Four Molecules Promise</h3>
<p><strong>Two Different Bets:</strong> NMN and NR are NAD boosters. They aim to raise NAD+, a molecule your cells use for energy that naturally declines with age. Resveratrol and metformin take a different angle, attempting to mimic some of the effects of eating less without the diet.</p>
<p>The theory is elegant. NAD+ does fall as we age, and animals on lifelong calorie restriction do tend to live longer and healthier. Borrowing those signals in a capsule is a reasonable hypothesis worth studying.</p>
<p>The question is whether raising a number in your blood, or copying a metabolic signal, actually adds healthy years to a human life. That leap from plausible mechanism to proven benefit is exactly where the marketing and the data part ways.</p>
<h3>What NMN and NR Actually Do</h3>
<p><strong>The Number Goes Up:</strong> Human trials confirm that oral NMN and NR are well tolerated and do raise NAD+ levels in the blood. That part is real and reproducible.</p>
<p>The harder question is what that higher number buys you. In aging mice, a related NAD precursor improved some measures of health but did not increase lifespan (<a href="https://www.nia.nih.gov/news/nicotinamide-supplement-aging-mice-shows-some-health-improvements-not-increased-longevity" target="_blank" rel="noopener">NIA, 2024</a>). In humans, the evidence for slowing aging itself remains thin and short-term.</p>
<p>Most human studies so far have run for weeks or a few months and enrolled small numbers of people. They were designed to check safety and confirm that NAD+ rises, not to follow people for the years it would take to measure aging itself.</p>
<p>If you are in your 40s or beyond and curious, the short-term safety record is reassuring. The anti-aging payoff is the part still waiting on the long, expensive trials that have not yet been done.</p>
<h3>The Resveratrol Reality Check</h3>
<p><strong>A Famous Compound With a Catch:</strong> Resveratrol, the polyphenol in red wine, may help protect blood vessels and modestly lower LDL cholesterol. But study results are genuinely mixed, with some showing benefit and others showing none.</p>
<p>There is also an absorption problem. Research suggests the body cannot take up most of the resveratrol in supplements, so a large share of what you swallow never reaches your bloodstream (<a href="https://www.mayoclinic.org/diseases-conditions/heart-disease/in-depth/red-wine/art-20048281" target="_blank" rel="noopener">Mayo Clinic, 2024</a>). You would also have to drink an impractical amount of wine to match the doses used in studies.</p>
<p>The more reliable move is food. Grapes, blueberries, cranberries, and peanuts all deliver resveratrol along with fiber and other compounds, and without the alcohol that undercuts the supposed heart benefit in the first place.</p>
<h3>Metformin's Open Longevity Question</h3>
<p><strong>A Diabetes Drug on Trial:</strong> Metformin has decades of safe use for type 2 diabetes, and animal studies hint it may slow some features of aging. That is why researchers have proposed formal human longevity trials.</p>
<p>Those trials are designed to test whether metformin can delay age-related diseases as a group, not just diabetes. They are ambitious, and they are not finished. Until they report, the longevity case rests largely on animal data and on observations in people who already have diabetes.</p>
<p>Metformin is not approved to extend lifespan in healthy people, and it is a prescription drug with real side effects, including digestive upset and, rarely, B12 depletion. Taking it off-label for longevity is a decision that belongs with a physician, not a supplement aisle.</p>
<h3>Spending Wisely on Longevity Pills</h3>
<p><strong>Foundations First:</strong> Dietary supplements are not reviewed by the FDA for effectiveness before they reach shelves, and natural does not mean risk-free.
